  The problem is I only know few days ago that the bell is similar to the one used in B9TL.. There are other buses fitted with these new bell, SBS 3723D SBS 276Y SBS 3728R.

  Route familiarisation could be also for new / current drivers to learn routes that they never drove before. Examples could be upcoming SP drivers or those who are transferring to drive another route.
  
  It's not necessary that route familiarisation means new service.

  I thought braking sounds for the Voith MBO405s are about the same for that of braking sounds of Voith Habit bendies?

  Yes Oceane... cos of the way it's being brought to idle position.
  
  For bus555, no... it doesn't have a different engine, nor a difference in engine sound. It's just that the Voith transmission made it sound funny. As i've taken SBS557L for some number of years, it has been a known fact that it sounds rather funny.
