Not necessarily. SMRT can just add 1 more train to the line to maintain the current frequency.

MRT's Boon Lay extension to start operating from 28 Feb 2009
SINGAPORE: The Boon Lay extension of the Mass Rapid Transit system will start operating from February 28 next year.
The 3.8-kilometre stretch along the East-West Line will consist of two stations - Pioneer and Joo Koon.
The Land Transport Authority says this will benefit Jurong West
residents as well as those working in the Jurong Industrial estate.
Their travelling time is expected to be cut by as much as 15 minutes.
The extension will also give commuters direct access to the MRT system.
They currently have to make bus transfers at Boon Lay station.

Not possible although NSL / EWL train is on automatic mode. If you want all train to be entirely driverless, the cost to upgrade the existing infrastructure to accomodate such major changes is very hefty. More and more computers are required for controlling driverless train without the driver onboard.
